Biological Background: macroH2A.1 Function and Localization

  • macroH2A.1 is a variant histone H2A that replaces conventional H2A in a subset of nucleosomes, where it represses transcription and contributes to nucleosome-mediated DNA compaction, limiting DNA accessibility to cellular machineries. Related references: PMID:12718888, PMID:15621527, PMID:16428466, PMID:15897469
  • Involved in stable X chromosome inactivation and inhibits the binding of transcription factors, including NF-kappa-B, while interfering with SWI/SNF remodeling complexes. Related references: PMID:15897469, PMID:12718888, PMID:16428466
  • Recruits class I HDACs and inhibits histone acetylation by EP300, inducing a hypoacetylated state of chromatin. Related references: PMID:16107708, PMID:16428466
  • Isoform 1 specifically binds poly-ADP-ribose and O-acetyl-ADP-ribose, playing a key role in NAD+ metabolism by capping PARP1-generated ADP-ribose chains, thereby limiting NAD+ consumption and supporting mitochondrial respiration. It also increases expression of redox genes such as SOD3. Related references: PMID:15902274, PMID:23022728
  • Isoform 2 does not bind poly-ADP-ribose and represses SOD3 gene expression. Related references: PMID:15902274, PMID:23022728
  • Localized to the nucleus and chromosomes.
  • Widely expressed across tissues.

Experimental Guidance and Technical Tips

  • The antibody was generated against a peptide spanning amino acids 200–300, which includes the non-histone macro domain. Consider this region's conservation when assessing cross-reactivity with other macroH2A variants.
  • For Western blotting, the predicted molecular weight is ~39 kDa. Due to the basic nature of histones, effective extraction may require specialized histone extraction protocols or acid-urea gels.
  • Validate the antibody in the relevant sample systems (human, mouse, rat) for IHC-P, IF/ICC, or ELISA as appropriate, using appropriate controls to confirm specificity.
