Biological Background: LZTS2 Function and Localization
- Leucine zipper putative tumor suppressor 2 (LZTS2), also known as LAPSER1, is a coiled-coil protein with a molecular weight of approximately 73 kDa.
- Functions as a negative regulator of katanin-mediated microtubule severing and release from the centrosome, and is required for central spindle formation and completion of cytokinesis.
- May negatively regulate axonal outgrowth by preventing the formation of microtubule bundles necessary for transport within the elongating axon.
- Acts as a negative regulator of the Wnt signaling pathway; represses beta-catenin-mediated transcriptional activation by promoting nuclear exclusion of beta-catenin.
- Predominantly localizes to the cytoplasm, cytoskeleton, and microtubule organizing center (centrosome).
- Highly expressed in prostate and testis, with slightly lower levels in spleen, thymus, uterus, small intestine, and colon.
- Associated with key cellular processes including cell cycle, cell division, and mitosis, and is characterized as a phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (amino acids 470–669) of human LZTS2, which may allow detection of full-length and relevant isoforms; consider sequence alignment to verify isoform coverage.
- For western blotting, standard protocols can be employed; a band near 73 kDa is expected. Validate in cell lines or tissues with confirmed LZTS2 expression (e.g., prostate, testis, or relevant cancer cell models).
- ELISA applications are suitable; ensure the recombinant protein or peptide coating buffers and assay conditions are optimized for this antibody.
- As the antibody cross-reacts with mouse and rat, it may be useful in comparative studies; however, confirm specificity in the respective species using positive and negative controls.
