Biological Background: LysRS/KARS Function and Localization
- Catalyzes the two-step attachment of lysine to tRNA(Lys): activation of lysine by ATP to form Lys-AMP, followed by transfer to the 3'-end of tRNA(Lys), a fundamental step in cytoplasmic protein synthesis. Related references: PMID:18029264, PMID:18272479, PMID:9278442
- Upon secretion, acts as a signaling molecule that triggers immune responses by activating monocytes/macrophages (PubMed:15851690). Related references: PMID:15851690

Experimental Guidance and Technical Tips
- The immunogen was designed using a recombinant protein spanning amino acids 298-597 of human LysRS. This region covers the C-terminal catalytic domain; thus, the antibody may recognize full-length and certain truncated isoforms if the epitope is preserved.
- For WB, a band around 68 kDa is expected, consistent with the calculated molecular weight. Consider optimizing blocking and detection conditions for the sample type and species.
- In IF/ICC, appropriate fixation and permeabilization methods should be used to access different subcellular pools (cytoplasmic, nuclear, mitochondrial).
- For ELISA, the antibody can be used in combination with appropriate capture/detection reagents; titration is recommended for each assay format.
- Cross-reactivity has been reported with human, mouse, and rat samples, but sequence homology suggests potential reactivity in other species; validate in the relevant experimental system.
