Biological Background: Lyn Function and Localization

  • Protein identity and isoforms: Tyrosine-protein kinase Lyn, encoded by the LYN (JTK8) gene, is a non-receptor tyrosine kinase. Alternative splicing produces isoforms p53Lyn and p56Lyn.
  • Core signaling function: Lyn transduces signals from cell surface receptors, acting as both a negative regulator and activator depending on context. It is critical for innate and adaptive immune responses, hematopoiesis, growth factor/cytokine signaling, integrin signaling, and responses to DNA damage.
  • B-cell regulation: Lyn is essential for B-cell receptor signaling, controlling initiation, down-regulation, differentiation, proliferation, survival, apoptosis, and immune self-tolerance.
  • Receptor coupling: It operates downstream of numerous immune receptors (BCR, CD79A/B, CD5, CD19, CD22, FCER1, FCGR2, TLR2, TLR4) and cytokine/chemokine receptors (EPOR, KIT, MPL, CXCR4, IL3/5, CSF2).
  • Endothelial function: Lyn regulates endothelial activation, neutrophil adhesion, and transendothelial migration. Related references: PMID:36932076
  • Downstream phosphorylation: Lyn phosphorylates ITIM motifs to recruit phosphatases (SHP-1, SHP-2, SHIP-1) that modulate signaling. It also directly phosphorylates BTK, CBL, CD5, CD19, SYK, TEC, BCAR1/CAS, and NEDD9/HEF1. Related references: PMID:9020138
  • Subcellular localization: Found at the cell membrane, nucleus, cytoplasm, perinuclear region, and Golgi apparatus. Membrane association is mediated by N-terminal myristoylation and palmitoylation.
  • Tissue specificity: Widely expressed in hematopoietic, neuronal, and epidermoid cells. Detected in monocytes, placenta, fetal brain, lung, liver, kidney, and primary neuroblastoma tumors.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-240) of human Lyn, covering both the unique and SH3 domains; this region is well conserved across human, mouse, and rat, consistent with the stated cross-reactivity.
  • For Western blot (WB), a band around 59 kDa is expected for the full-length protein, with possible detection of splice isoforms p53 and p56 at lower molecular weights. Validate in your specific lysate type.
  • In ELISA, consider using the antibody as a capture or detection reagent, but optimal dilution should be determined experimentally.
  • Lyn is involved in B-cell receptor signaling; therefore, stimulation with anti-IgM or pervanadate may be required to detect activation-dependent phosphorylation events in downstream targets.
