Biological Background: LY96 Function and Localization
-
LY96 (lymphocyte antigen 96), also known as MD-2 or ESOP-1, binds bacterial lipopolysaccharide (LPS) and cooperates with Toll-like receptor 4 (TLR4) to mediate innate immune responses, and cells expressing both LY96 and TLR4, but not TLR4 alone, respond to LPS. Related references: PMID:17569869, PMID:17803912, PMID:11160242, PMID:11593030, PMID:10359581
-
LY96 also cooperates with TLR2 in the response to cell wall components from Gram-positive and Gram-negative bacteria. Related references: PMID:11160242, PMID:11593030
-
LY96 enhances TLR4-dependent activation of NF-kappa-B, a key transcription factor in inflammatory signaling. Related references: PMID:10359581
-
The protein is secreted and found in the extracellular space, consistent with its role as a soluble co-receptor.
-
LY96 undergoes alternative splicing, contains disulfide bonds, and is glycosylated, as indicated by keywords from curated databases.
-
It is classified under immunity, inflammatory response, and innate immunity, reflecting its central function in pathogen recognition.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 19–160, the mature secreted form; the antibody is expected to recognize both native and recombinant LY96.
- For western blotting, a band of approximately 19 kDa is predicted; consider using reducing sample treatment, but note that disulfide bonds may affect migration under non-reducing conditions.
- Validate antibody specificity in the relevant biological context (e.g., LPS-stimulated vs. unstimulated cells) and include appropriate controls.
- For ELISA, this antibody can be used as a capture or detection reagent; pairing with another MD‑2 antibody may improve sensitivity in sandwich assays.
- Confirmed cross-reactivity with mouse enables analysis in mouse-derived samples and models.
- Because LY96 is secreted, cell culture supernatants or biological fluids (e.g., serum) are suitable sample types.
