Biological Background: LXRα Function and Localization
- LXRα (Oxysterols receptor LXR-alpha) is a ligand-activated nuclear receptor and transcription factor that regulates gene expression in response to oxysterols. Related references: PMID:19481530 PMID:25661920 PMID:37478846
- Upon activation, LXRα forms a heterodimer with the retinoic acid receptor (RXR), converting RXR from a silent partner into an active ligand-binding subunit that mediates retinoid responses through LXRE response elements. Related references: PMID:37478846
- LXRα is a central regulator of cholesterol homeostasis, controlling cholesterol uptake via MYLIP-dependent ubiquitination of LDLR, VLDLR, and LRP8 receptors. Related references: PMID:19481530
- It functionally interacts with RORA to co-regulate hepatic metabolic genes.
- LXRα induces LPCAT3-dependent phospholipid remodeling in the endoplasmic reticulum of hepatocytes, driving SREBF1 processing and de novo lipogenesis.
- Through LPCAT3, LXRα promotes arachidonate incorporation into phosphatidylcholines, increasing ER membrane fluidity and facilitating VLDL triacylglycerol transfer.
- LXRα also mitigates lipid-induced ER stress and inflammation by modulating SRC kinase compartmentalization and limiting pro-inflammatory lipid mediator synthesis.
- Predominantly localized in the nucleus with some cytoplasmic presence, LXRα is strongly expressed in visceral organs including liver, kidney, intestine, and spleen. It contains conserved zinc-finger DNA-binding domains typical of nuclear receptors.
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 160–230 of human LXRα; this region may provide specificity for LXRα over other nuclear receptors. Validate the antibody’s performance with your target isoform and sample type.
- For Western blot, consider using nuclear-enriched lysates from liver or kidney tissue, as LXRα is highly expressed in these organs. A band at ~50 kDa is expected; optimization of blocking and antibody dilution may be required.
- In ELISA, the antibody can serve as a capture or detection reagent for recombinant LXRα or endogenous LXRα in cell/tissue extracts. Pair with a suitable secondary antibody and validate with known standards.
- Cross-reactivity with mouse and rat LXRα allows potential use in rodent models; however, confirm reactivity in your specific experimental system.
- Since LXRα is a ligand-activated transcription factor, consider including appropriate controls (e.g., ligand treatment) when assessing expression changes.
