Biological Background: LXR beta/NER Function and Localization
- Also known as Liver X receptor beta, Nuclear receptor NER, NR1H2, and RIP15 (Gene ID: 7376). Encodes a ~51 kDa nuclear receptor protein.
- Functions as a ligand-dependent transcriptional activator, binding to DNA elements with the consensus sequence 5'-AGGTCA-3' (DR-4) (PubMed:25661920). Related references: PMID:25661920
- Regulates cholesterol homeostasis by mediating MYLIP-dependent ubiquitination of LDLR, VLDLR, and LRP8, thereby controlling lipoprotein uptake.
- Interplays with RORA to regulate liver metabolic genes and induces LPCAT3-dependent phospholipid remodeling in ER membranes, driving lipogenesis and VLDL assembly.
- Acts as an anti-inflammatory corepressor during the hepatic acute phase response by preventing N-Cor corepressor complex dissociation (PubMed:20159957). Related references: PMID:20159957
- Localizes to the nucleus and is ubiquitously expressed across tissues.
- Belongs to the nuclear receptor superfamily, featuring zinc-finger DNA-binding domains and ligand-binding domains; undergoes Ubl conjugation and isopeptide bond formation.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide spanning amino acids 180-260 of human LXR beta/NER. For WB, consider using whole cell lysates or nuclear extracts.
- A band near 51 kDa is expected. Optimize blocking and antibody dilution empirically using lysates from human, mouse, or rat tissues known to express LXR beta (e.g., liver, kidney).
- For IF/ICC, fixation and permeabilization conditions are critical for nuclear protein detection. Use appropriate controls and validate signal specificity.
- In ELISA, the antibody can serve as a capture or detection reagent; pairing with a matched monoclonal antibody may enhance sensitivity.
