Biological Background: Lactotransferrin (LTF) Function and Localization
- Lactotransferrin (LTF) is a major iron-binding glycoprotein belonging to the transferrin family, which can bind two Fe³⁺ ions in association with bicarbonate (PubMed:11179314, PubMed:12693969, PubMed:14573629). Related references: PMID:11179314 PMID:12693969 PMID:14573629
- It is highly abundant in exocrine fluids such as breast milk, tears, and saliva, and plays a critical role in innate immunity through both iron sequestration and direct antimicrobial activity against bacteria, fungi, and biofilms (PubMed:6802759, PubMed:12037568, PubMed:11083624). Related references: PMID:6802759 PMID:12037568 PMID:11083624
- LTF modulates inflammatory responses by stimulating TLR4 signaling and NF-kB activation, while also interfering with LPS-stimulated TLR4 pathways, and inhibits neutrophil migration to apoptotic sites (PubMed:20345905, PubMed:19033648). Related references: PMID:20345905 PMID:19033648
- It promotes cell proliferation and differentiation, showing anabolic effects on osteoblasts, stimulation of VEGF-mediated endothelial cell migration, and anti-apoptotic activity (PubMed:15166119, PubMed:16842782). Related references: PMID:15166119 PMID:16842782
- LTF binds a variety of molecules including heparin, chondroitin sulfate, bacterial lipopolysaccharide, and DNA, which may contribute to its antimicrobial and immunomodulatory functions (PubMed:9359845). Related references: PMID:9359845
- The transferrin-like domain 1 exhibits serine protease activity (peptidase S60 family), cleaving arginine-rich sequences, which enhances its antimicrobial properties (PubMed:12535064). Related references: PMID:12535064
- A truncated isoform, DeltaLf, acts as a transcription factor, binding to promoter elements of genes like SKP1 and BAX to induce cell cycle arrest (PubMed:15222485, PubMed:22320386). Related references: PMID:15222485 PMID:22320386
- Tissue expression is broad: high in saliva/tears, moderate in serum/plasma, low in urine; detected in distal collecting tubules of kidney and peripheral blood neutrophils. Isoform DeltaLf is expressed in placenta, liver, ovary, while full-length LTF is abundant in kidney medulla, brain, and leukocytes.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant protein spanning amino acids 21–352 of human LTF. For Western blot, the predicted molecular weight is ~78 kDa; consider testing whole cell lysates from appropriate tissues (e.g., mammary, salivary) or secreted fractions given LTF’s exocrine nature, and validate in your sample system.
- For immunohistochemistry (IHC-P), appropriate fixation and antigen retrieval conditions should be empirically determined. LTF is highly expressed in breast, kidney medulla, and salivary glands, which can serve as positive tissue controls.
- In ELISA applications, the antibody can be used for detection of LTF in biological fluids like milk, tears, or serum; consider pairing with a matched detection system and standard curve.
- Due to its polyclonal nature, lot-to-lot variation may occur; inclusion of appropriate controls and validation within your specific experimental context is recommended.
