Biological Background: LTBR Function and Localization
- LTBR (TNFRSF3) is a member of the tumor necrosis factor receptor superfamily and is also known as lymphotoxin-beta receptor, TNFCR, TNFR3, TNFR-RP, TNFRSF3, TNFR2-RP, LT-BETA-R, and TNF-R-III.
- It functions as a receptor for heterotrimeric lymphotoxin (composed of LTA and LTB) and for TNFS14/LIGHT. Ligand binding activates the NF-kappa-B signaling pathway. Related references: PMID:24248355
- Through adaptor proteins TRAF3 and TRAF5, LTBR promotes apoptosis.
- It may play a role in the development of lymphoid organs.
- LTBR is localized to the plasma membrane.
- The protein undergoes post-translational modifications including disulfide bond formation, glycosylation, and phosphorylation.
- Alternative splicing of the LTBR gene results in multiple transcript variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the intracellular C-terminal region (aa 336-435) of human LTBR, ensuring specific detection of endogenous receptor.
- For western blot (WB), the predicted molecular weight is 47kDa; however, the protein may exhibit altered migration due to glycosylation.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), membrane staining is expected; appropriate antigen retrieval and controls should be optimized.
- Cross-reactivity with mouse and rat LTBR supports comparative studies; validate using species-appropriate lysates or tissues.
- The antibody is suitable for ELISA-based detection; titrate antibody concentration according to plate format and assay conditions.
- Perform standard blocking and antibody diluent optimization to reduce nonspecific binding in these applications.
