Biological Background: LTB4R Function and Localization

  • LTB4R (leukotriene B4 receptor 1) is also known as BLT1, BLTR, P2Y7, GPR16, CMKRL1, and LTBR1. It is a multi-pass membrane protein belonging to the G protein-coupled receptor (GPCR) family.
  • It functions as a high-affinity receptor for leukotriene B4 (LTB4), a potent lipid chemoattractant involved in inflammation and immune responses. Activation of the receptor triggers G protein-mediated signaling cascades that elevate intracellular calcium.
  • Besides LTB4, this receptor also responds to extracellular nucleotides, with a preference for ATP over UTP and ADP, suggesting a dual role in purinergic and leukotriene signaling.
  • The receptor is primarily localized at the cell membrane, consistent with its role in transmitting extracellular signals into the cell.
  • According to UniProt keywords, LTB4R undergoes post-translational modifications including phosphorylation and glycosylation, which may regulate receptor function and trafficking.
  • LTB4R is expressed at highest levels in heart and skeletal muscle, with lower levels in brain and liver. It is also highly expressed in lymphoid tissues, underscoring its role in immune cell chemotaxis and inflammation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a synthetic peptide from the C-terminal intracellular domain of human LTB4R (residues 253–352). This region is less conserved across species, which is consistent with the reported cross-reactivity to mouse and rat.
  • In WB, a band at approximately 38 kDa is expected; however, due to glycosylation, the observed molecular weight may be slightly higher. Always validate in the relevant sample system using positive controls.
  • For ELISA, consider pairing this antibody with a matched detection reagent if developing a sandwich assay. Since the antibody is polyclonal, lot-to-lot consistency should be monitored.
  • Given its cross-reactivity with mouse and rat, this antibody can be used to detect LTB4R in rodent tissues, but preliminary specificity testing in those species is recommended.
