Biological Background: Leukotriene A-4 hydrolase Function and Localization

  • Leukotriene A-4 hydrolase (LTA4H) is a bifunctional zinc metalloenzyme possessing both epoxide hydrolase and aminopeptidase activities (UniProt P09960).
  • The epoxide hydrolase activity catalyzes the conversion of leukotriene A4 (LTA4) to the pro-inflammatory mediator leukotriene B4 (LTB4). Related references: PMID:11917124, PMID:12207002, PMID:15078870
  • LTA4H can also use LTA5 as a substrate, albeit less efficiently, to produce LTB5. Related references: PMID:2996528
  • The aminopeptidase activity displays high affinity for N-terminal arginines of tripeptides and inactivates the neutrophil chemoattractant Pro-Gly-Pro (PGP) by cleavage, thus counteracting inflammation. Related references: PMID:18804029, PMID:20813919, PMID:24591641
  • LTA4H is involved in the biosynthesis of resolvin E1 and 18S-resolvin E1 from eicosapentaenoic acid, which are lipid mediators with potent anti-inflammatory and pro-resolving actions. Related references: PMID:21206090
  • The enzyme localizes to the cytoplasm and is expressed across multiple blood cell types, including monocytes, lymphocytes, neutrophils, reticulocytes, platelets, and fibroblasts, in both isoform 1 and isoform 2.
  • Key post-translational modifications include acetylation and phosphorylation, and the protein is a zinc-dependent metalloprotease.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to amino acids 312–611 of human LTA4H (NP_000886.1), covering the C-terminal region. This region includes the catalytic domain and may recognize both the full-length protein and potential isoforms.
  • For Western blot (WB), the predicted molecular weight is ~69 kDa. Consider using whole cell lysates from human or mouse leukocyte-rich samples (e.g., peripheral blood mononuclear cells) or cell lines such as THP-1 or RAW264.7, which are known to express LTA4H.
  • For ELISA, the antibody can be used as a capture or detection reagent after in-house validation. Pair it with a matched monoclonal antibody if available for sandwich ELISA to detect soluble LTA4H in serum or plasma.
  • Since cross-reactivity is listed for human and mouse, verify reactivity with your specific sample type and consider using blocking peptides to confirm specificity.
