Biological Background: LTA4H Function and Localization
- LTA4H is a bifunctional zinc metalloenzyme possessing both epoxide hydrolase and aminopeptidase activities.
- Acts as an epoxide hydrolase to convert LTA4 into the pro-inflammatory mediator LTB4. Can also utilize LTA5 as a substrate, though less efficiently, to generate LTB5. Related references: PMID:11917124 PMID:12207002 PMID:15078870 PMID:2996528
- Functions as an aminopeptidase with high affinity for N-terminal arginines of tripeptides, and inactivates the neutrophil chemoattractant Pro-Gly-Pro (PGP), potentially providing an anti-inflammatory balance. Related references: PMID:18804029 PMID:20813919 PMID:24591641
- Participates in the biosynthesis of resolvin E1 and 18S-resolvin E1 from eicosapentaenoic acid, contributing to the resolution of inflammation. Related references: PMID:21206090
- Predominantly localized in the cytoplasm.
- Expressed as isoform 1 and isoform 2 in monocytes, lymphocytes, neutrophils, reticulocytes, platelets, and fibroblasts.
- Zinc-dependent catalysis; post-translationally modified by acetylation and phosphorylation.
- Also designated as Leukotriene A(4) hydrolase and Tripeptide aminopeptidase LTA4H.
Experimental Guidance and Technical Tips
- This antibody was generated using a recombinant immunogen covering amino acids 501–600 of human LTA4H, a region within the C-terminal domain; this region is conserved in mouse and rat, consistent with the reported cross-reactivity.
- For Western blotting, validate the antibody with appropriate positive controls (e.g., leukocyte or platelet lysates) and optimize blocking and antibody dilution for your specific sample type.
- In immunofluorescence and ICC, confirm LTA4H expression in cell lines known to express the target, and consider proper fixation/permeabilization protocols (e.g., methanol or paraformaldehyde) to preserve the epitope.
- For ELISA applications, this antibody may be used as a capture or detection reagent; empirical determination of optimal coating concentrations and pairing antibodies is recommended.
