Biological Background: LSM1 Function and Localization
- LSM1, also known as Cancer-associated Sm-like (CaSm) or CASM, is a U6 snRNA-associated Sm-like protein.
- Plays a critical role in the degradation of histone mRNAs, the only eukaryotic mRNAs that are not polyadenylated. Related references: PMID:18172165
- Functions as part of an LSm subunit-containing complex that participates in the general process of cytoplasmic mRNA decay (By similarity).
- Localizes primarily to the cytoplasm and is enriched in P-bodies, dynamic cytoplasmic foci involved in mRNA turnover.
- Belongs to the highly conserved Sm/LSm protein family, key components of ribonucleoprotein complexes involved in RNA processing.
- Phosphorylation has been reported, indicating potential regulatory mechanisms controlling its activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–133) of human LSM1, which may be effective for detecting both native and denatured protein forms.
- For Western blotting, expect a band at approximately 15 kDa; validate in the relevant human or mouse cell/tissue lysate.
- In immunofluorescence/ICC, anticipate cytoplasmic staining with possible enrichment in P-body-like foci; co-staining with P-body markers (e.g., DCP1A) can confirm localization.
- For ELISA, optimize coating antigen concentration and antibody dilution for best signal-to-noise ratio.
- Since this is a polyclonal antibody, consistency across lots can vary; consider performing lot-specific validation for critical experiments.
- Cross-reactivity with mouse allows analysis in rodent models; verify performance in the specific model system.
