Biological Background: Lsh/HELLS Function and Localization
- Lsh/HELLS is an ATP-dependent chromatin remodeler that facilitates de novo DNA methylation and recruits DNMTs/HDACs to promote transcriptional silencing, heterochromatin formation, and repression of transposable elements.
Related references: PMID:30307408 - It is recruited to double-strand breaks in heterochromatin, aiding DNA repair via homologous recombination by mediating RBBP8/CtIP-dependent end resection.
Related references: PMID:22946062 | PMID:31802118 - During meiosis, PRDM9 recruits HELLS to recombination hotspots to facilitate chromatin opening, enabling meiotic recombination.
Related references: PMID:32001511 - It is involved in the regulation of expansion or survival of lymphoid cells (By similarity).
- HELLS localizes to the nucleus and associates with chromosomes.
- Highly expressed in proliferative tissues such as thymus and testis, and in neoplastic cell lines; lower expression in uterus, small intestine, colon, and peripheral blood mononuclear cells.
- Contains ATP-binding and helicase domains, is subject to phosphorylation, and is linked to ICF4 syndrome (immunodeficiency-centromeric instability-facial anomalies syndrome 4).
Experimental Guidance and Technical Tips
- The immunogen (amino acids 614-838 of human HELLS, NP_060533.2) may be conserved across human, mouse, and rat, consistent with reported cross-reactivity. Validate the antibody in your specific sample system and assay conditions.
- This rabbit monoclonal antibody is validated for WB, IF/ICC, IF-P, IHC-P, and ELISA. For WB, expect a band at ~97 kDa; use appropriate molecular weight markers and positive/negative controls.
- Given HELLS's roles in chromatin remodeling, DNA repair, and epigenetic regulation, this antibody may be useful for studies of heterochromatin dynamics and genome stability. For IF/IHC, optimize fixation and permeabilization protocols based on your sample type.
