Biological Background: LRRK2 Function and Localization
- LRRK2 (Leucine-rich repeat serine/threonine-protein kinase 2), also known as Dardarin or PARK8, is a large multidomain protein possessing both serine/threonine kinase and GTPase activities; the GTPase function via the Roc domain regulates its kinase activity. Related references: PMID:18230735, PMID:26824392, PMID:28720718
- This kinase phosphorylates a broad array of substrates involved in critical cellular processes including neuronal plasticity, innate immunity, autophagy, and vesicle trafficking. Related references: PMID:17114044, PMID:20949042, PMID:21850687
- LRRK2 is a master regulator of RAB GTPases; it phosphorylates multiple RAB family members (RAB3, RAB5, RAB8, RAB10, RAB12, RAB29, RAB35, RAB43), thereby controlling their GDP/GTP exchange and interactions with effectors. Related references: PMID:23395371, PMID:26824392, PMID:28720718
- LRRK2 positively regulates autophagy via a calcium-dependent mechanism that activates the CaMKK/AMPK signaling pathway, involving lysosomal calcium release. Related references: PMID:22012985
- In neurons, LRRK2 modulates process morphology, synaptic vesicle trafficking, and primary ciliogenesis; it phosphorylates RAB8A and RAB10 to promote sonic hedgehog (SHH) signaling in the brain. Related references: PMID:17114044, PMID:24687852, PMID:29125462, PMID:30398148
- Mutations in the LRRK2 gene are the most common genetic cause of familial Parkinson’s disease (PARK8) and also contribute to sporadic cases.
- Subcellularly, LRRK2 localizes to numerous membrane compartments including the Golgi apparatus, endoplasmic reticulum, endosomes, lysosomes, mitochondria, and synaptic vesicles; it is also found in the cytosol and associated with the cytoskeleton.
- LRRK2 is broadly expressed, with high levels in brain, heart, and liver; in the central nervous system, it is prominent in dopaminoceptive areas and cortical pyramidal neurons.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 100–340 of human LRRK2, an N-terminal region; consider epitope accessibility in native versus denaturing conditions when designing detection assays.
- For Western blotting, use low-percentage SDS-PAGE gels and extended transfer times suitable for high-molecular-weight proteins (~286 kDa).
- In ELISA applications, validate detection using recombinant LRRK2 protein or lysates from cell lines that express the target endogenously.
- Cross-reactivity is confirmed for human; for other species, assess sequence homology of the immunogen region and perform a pilot experiment.
