Biological Background: LRRFIP1 Function and Localization
- LRRFIP1 (Leucine-rich repeat flightless-interacting protein 1), also known as GCF2 and TRIP, is a transcriptional repressor that preferentially binds to GC-rich consensus sequences to regulate expression of TNF, EGFR, and PDGFA.
- It may control smooth muscle cell proliferation following arterial injury through PDGFA repression.
- LRRFIP1 binds double-stranded RNA and positively regulates Toll-like receptor (TLR) signaling by competing with the negative regulator FLII for MYD88-binding.
- The protein localizes to both the nucleus and cytoplasm, consistent with its roles in transcription and cytoplasmic signaling.
- It is ubiquitously expressed across tissues, reflecting its broad regulatory functions.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin-like conjugation; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 589–809) resides in the C-terminal region of LRRFIP1. Given the high conservation among mammals, the antibody is likely to recognize the endogenous protein in human, mouse, and rat samples.
- For Western blot, validate detection in whole-cell lysates or nuclear extracts from relevant cell lines; the predicted molecular weight is ~89 kDa, but phosphorylation or other modifications may cause slight mass shifts.
- For ELISA, consider using the antibody as a capture or detection reagent following standard protocols; specific titration under your experimental conditions is recommended.
- As LRRFIP1 is a transcriptional regulator, enrichment of nuclear fractions can enhance signal intensity in WB.
