Biological Background: LRPPRC Function and Localization
- LRPPRC (Leucine-rich PPR motif-containing protein) is a large 130–158 kDa protein that localizes to both mitochondria and the nucleus, where it participates in RNA metabolism, transcription regulation, and mitochondrial function.
- In the nucleus, LRPPRC binds to HNRPA1-associated poly(A) mRNAs and positively modulates nuclear export of 4ESE-containing mRNAs by interacting with EIF4E and exportin XPO1/CRM1, assembling nuclear mRNA ribonucleoprotein complexes. Related references: PMID:19262567 PMID:28325843
- In mitochondria, LRPPRC binds poly(A) mRNA and is required for the translation or stability of mitochondrially encoded cytochrome c oxidase (COX) subunits; it also cooperates with PPARGC1A to regulate transcription of certain mitochondrial and gluconeogenic genes.
- LRPPRC is essential for maintaining mitochondrial potential and suppresses basal autophagy and mitophagy by sustaining BCL2 levels. Related references: PMID:23822101
- The protein is expressed ubiquitously, with highest levels in heart, skeletal muscle, kidney, and liver; it is associated with primary mitochondrial disease and Leigh syndrome (MC4DN5).
- LRPPRC undergoes post-translational modifications such as acetylation and phosphorylation, and contains a transit peptide for mitochondrial targeting.
Experimental Guidance and Technical Tips
- Due to its large molecular weight (158 kDa), consider using a high-range molecular weight marker and extended transfer times for Western blot analysis.
- For IHC-P and IF/ICC, validate the antibody in tissues with known high LRPPRC expression (heart, skeletal muscle, liver, kidney) as positive controls; expect mitochondrial and nuclear staining.
- The immunogen spans amino acids 1041–1394, which may influence detection of specific isoforms; for cross-reactivity testing, include appropriate human, mouse, and rat lysates.
- For ELISA, titrate the antibody against the recombinant immunogen or a relevant full-length protein standard.
