Biological Background: LRP6 Function and Localization
- LRP6 (Low-density lipoprotein receptor-related protein 6), also known as ADCAD2 or STHAG7, is a cell-surface coreceptor of the canonical Wnt/β-catenin signaling pathway. It forms the Wnt-Fzd-LRP5-LRP6 complex that triggers β-catenin signaling by inducing the aggregation of receptor-ligand complexes into ribosome-sized signalosomes (PubMed:11357136, PubMed:11448771, PubMed:15778503). - Upon Wnt stimulation, LRP6 recruits DVL1 polymers and the AXIN1/GSK3B destruction complex to the plasma membrane. This inhibits AXIN1/GSK3-mediated phosphorylation and proteasomal degradation of β-catenin, thereby stabilizing it for nuclear translocation and target gene activation (PubMed:16513652).
