Biological Background: LRP5 Function and Localization
- LRP5, also known as LRP-7, is a single-pass type I membrane protein and a member of the low-density lipoprotein receptor family.
- LRP5 acts as a coreceptor with Frizzled family proteins to transduce Wnt signals and activate the canonical Wnt/β-catenin pathway, regulating cell fate determination, self-renewal, and embryonic development. Related references: PMID:11336703, PMID:11448771, PMID:11719191
- During bone development, LRP5 regulates osteoblast proliferation and differentiation, thereby determining bone mass. Related references: PMID:11719191
- Mechanistically, the formation of the Wnt-Frizzled-LRP5 signaling complex promotes AXIN1 recruitment, stabilizing β-catenin and activating TCF/LEF transcriptional programs. Related references: PMID:11336703, PMID:14731402, PMID:24706814
