Biological Background: LRP4 Function and Localization

  • LRP4 (Low-density lipoprotein receptor-related protein 4) is a transmembrane receptor encoded by the LRP4 gene. It is also known as MEGF7 (Multiple epidermal growth factor-like domains 7), LRP10, and SOST2, and belongs to the LDL receptor family.
  • LRP4 mediates SOST-dependent inhibition of bone formation by functioning as a specific facilitator of SOST-mediated inhibition of Wnt signaling.
  • It plays a crucial role in the formation and maintenance of the neuromuscular junction (NMJ) by directly binding AGRIN and recruiting it to the MUSK signaling complex, which induces MUSK phosphorylation and subsequent clustering of acetylcholine receptors (AChR) in the postsynaptic membrane.
  • LRP4 may also negatively regulate the canonical Wnt signaling pathway by antagonizing LRP6-mediated activation.
  • It has been proposed to function as a cell surface endocytic receptor that binds and internalizes extracellular ligands for degradation by lysosomes.

  • Subcellular location: Predominantly localized to the cell membrane.
