Biological Background: LRAT Function and Localization
- LRAT (Lecithin retinol acyltransferase, also known as phosphatidylcholine--retinol O-acyltransferase) is a 25.7 kDa membrane-associated enzyme that transfers an acyl group from phosphatidylcholine to all-trans retinol, yielding all-trans retinyl esters, the primary storage form of vitamin A. - These retinyl esters are essential for vision, serving as substrates for the isomerohydrolase in the retinal pigment epithelium, which generates 11-cis-retinol that is subsequently oxidized to 11-cis-retinaldehyde—the chromophore for rhodopsin and cone photopigments.
- LRAT is required for the survival of cone photoreceptors and the maintenance of normal rod photoreceptor cell morphology.
- The protein localizes primarily to the endoplasmic reticulum membrane and rough ER, and is also detected in endosomes, multivesicular bodies, and the cytoplasmic perinuclear region.
- At the protein level, LRAT is highly expressed in testis and liver, with significant levels also in the retinal pigment epithelium, small intestine, prostate, pancreas, and colon; low expression is observed in the brain.
