Biological Background: LPAR2 Function and Localization

  • LPAR2 (Lysophosphatidic acid receptor 2), also known as EDG-4 or LPA2, is a G protein-coupled receptor that binds lysophosphatidic acid (LPA), a bioactive lipid mediator involved in many cellular processes.
  • It couples to multiple G protein families, including G(i)/G(o), G(12)/G(13), and G(q), thereby activating diverse downstream signaling cascades.
  • The receptor plays a key role in the phospholipase C-beta (PLC-beta) signaling pathway, stimulating phospholipase C (PLC) activity in a manner independent of RALA activation.
  • LPAR2 is located at the cell surface and cell membrane, consistent with its function as a transmembrane receptor.
  • Tissue expression is highest in testes and peripheral blood leukocytes, with lower levels in pancreas, spleen, thymus, and prostate, and little to no expression in many other tissues.
  • Post-translational modifications include glycosylation, palmitoylation, and lipoprotein modifications, as indicated by keywords (Glycoprotein, Lipoprotein, Palmitate).
  • The receptor belongs to a family of lysophosphatidic acid receptors, with a molecular weight of approximately 39 kDa.

Experimental Guidance and Technical Tips

  • For Western blot (WB), consider validating the antibody in lysates from tissues with high LPAR2 expression, such as testes or peripheral blood leukocytes, or in cell lines known to express the receptor.
  • Use the immunizing peptide (amino acids 250-350) as a blocking control in ELISA or WB to confirm binding specificity.
  • Optimize antibody dilution and incubation conditions for each application, as performance may vary depending on sample type and assay setup.
  • When performing ELISA, ensure proper coating and detection systems are in place, and include appropriate negative controls to assess background.
