Biological Background: LOXL2 Function and Localization

  • LOXL2 (Lysyl oxidase homolog 2) is a copper-dependent amine oxidase that mediates oxidative deamination of lysine residues. It acts as a transcription corepressor by specifically deaminating trimethylated 'Lys-4' of histone H3 (H3K4me3) and also deaminates TAF10 to release it from promoters, inhibiting TFIID-dependent transcription. Related references: PMID:27735137 PMID:25959397
  • It shows strict substrate specificity, active only on H3K4me3 and not on H3K9me3, H3K27me3, or partially methylated H3K4, highlighting its precise role in epigenetic transcriptional regulation. Related references: PMID:27735137
  • LOXL2 is a key mediator of epithelial-to-mesenchymal transition (EMT). It interacts with SNAI1 to repress E-cadherin (CDH1) expression, likely through histone H3 deamination. LOXL2 also binds endoplasmic reticulum HSPA5, activating the IRE1-XBP1 pathway to drive EMT. Related references: PMID:16096638 PMID:24414204 PMID:27735137 PMID:28332555
  • Under hypoxic conditions, LOXL2 represses E-cadherin, contributing to tumor aggressiveness. It is frequently upregulated in various cancers, suggesting a role in tumor progression. Related references: PMID:20026874
  • Secreted LOXL2 functions in the extracellular matrix, crosslinking collagen and elastin through oxidative deamination of peptidyl lysines. It also regulates sprouting angiogenesis via collagen IV scaffolding. Related references: PMID:20306300 PMID:21835952
  • The enzyme contains a lysyl tyrosylquinone (LTQ) cofactor essential for its catalytic activity, formed by post-translational crosslinking of lysine and tyrosine residues. It also binds copper and features multiple disulfide bonds and glycosylation sites.
  • Subcellular localization is dual: LOXL2 is secreted to the extracellular matrix and basement membrane but also resides in the nucleus, chromosomes, and endoplasmic reticulum, consistent with its transcriptional regulatory and extracellular functions.
  • LOXL2 is broadly expressed, with highest levels in reproductive tissues (placenta, uterus, prostate). Expression is elevated in esophageal epithelium layers and numerous carcinomas, making it a potential biomarker. Related references: PMID:10212285 PMID:22204712

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 51–290, encompassing part of the LOX catalytic domain. This region may detect both full-length and truncated LOXL2 isoforms; validate antibody specificity in your target system.
  • For Western blot (WB), prepare lysates with protease/phosphatase inhibitors, and use a loading control compatible with the expected 87 kDa band. Confirm reactivity with human, mouse, and rat samples as listed.
  • In ELISA applications, consider pairing with a detection antibody that recognizes a non-overlapping LOXL2 epitope for sandwich assays. Optimize coating, blocking, and detection conditions empirically.
  • Because LOXL2 localizes to both nuclear and secreted compartments, select lysis buffers and fractionation methods appropriate for your sample type to ensure target recovery.
