Biological Background: LOXL2 Function and Localization

  • LOXL2 (Lysyl oxidase homolog 2), also known as LOR, LOR2, or WS9-14, is a member of the lysyl oxidase family and functions as an oxidoreductase that requires copper and LTQ cofactors.
  • Secreted LOXL2 promotes cross-linking of collagen and elastin precursors in the extracellular matrix, which is essential for matrix integrity and sprouting angiogenesis. Related references: PMID:20306300 PMID:21835952
  • In the nucleus, LOXL2 acts as a transcription corepressor by specifically deaminating trimethylated lysine 4 of histone H3 (H3K4me3), a mark of active transcription, leading to transcriptional repression. Related references: PMID:27735137
  • LOXL2 also mediates deamination of TAF10, a component of the TFIID complex, causing its release from promoters and inhibiting TFIID-dependent transcription, including repression of pluripotency genes like POU5F1/OCT4, NANOG, KLF4, and SOX2. Related references: PMID:25959397
  • During epithelial-to-mesenchymal transition (EMT), LOXL2 interacts with SNAI1 to repress E-cadherin (CDH1) expression via histone H3 deamination. It also is recruited to pericentromeric heterochromatin to oxidize histone H3, releasing CBX5/HP1A and enabling chromatin reorganization. Additionally, LOXL2 interacts with HSPA5 to activate the IRE1-XBP1 pathway of the unfolded protein response, further driving EMT. Related references: PMID:16096638 PMID:24414204 PMID:24239292 PMID:28332555
  • LOXL2 is localized to the extracellular space, extracellular matrix, basement membrane, nucleus, chromosome, and endoplasmic reticulum.
  • LOXL2 is widely expressed, with highest levels in reproductive tissues (placenta, uterus, prostate) and is upregulated in many cancers. In esophageal epithelium, it is detected in basal, prickle, and granular cell layers. Related references: PMID:10212285 PMID:22204712
  • Key features include calcium binding, copper binding, disulfide bond formation, glycoprotein, LTQ cofactor, metal-binding, and its role as a chromatin regulator, repressor, and oxidoreductase.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to the C-terminal region (amino acids 675-774) of human LOXL2, which may confer specificity against other lysyl oxidase family members. Researchers should verify cross-reactivity if needed.
  • For Western blot (WB), the predicted molecular weight is approximately 87 kDa. Post-translational modifications might cause slight shifts; validate with appropriate controls and sample preparation.
  • For ELISA, consider using recombinant LOXL2 protein or the immunizing peptide as a positive control. Optimize antibody dilution and assay conditions for the specific sample matrix.
  • Always include relevant positive and negative controls, and pre-validate the antibody in the target application using known standards.
