Biological Background: LONP2 Function and Localization
- LONP2 (Lon protease homolog 2, peroxisomal) is also known as Lon protease-like protein 2 (Lon protease 2) and peroxisomal Lon protease.
- It is an ATP-dependent serine protease that mediates selective degradation of misfolded and unassembled polypeptides in the peroxisomal matrix (By similarity).
- LONP2 is required for the processing of type 2 peroxisome targeting signal (PTS2)-containing proteins and facilitates peroxisome matrix protein import.
- It may indirectly regulate peroxisomal fatty acid β-oxidation by degrading self-processed forms of TYSND1.
- The protein localizes to the peroxisome matrix.
- It is widely expressed, with highest expression in liver, kidney, and pancreas.
- Post-translational modifications include acetylation; alternative splicing produces multiple isoforms.

- For Western blotting, use whole-cell lysates from human, mouse, or rat tissues (e.g., liver, kidney) or cell lines; note the predicted molecular weight of ~95 kDa (precursor) and consider possible alternatives due to processing.
- Optimize antibody dilution empirically; a starting range of 1:500–1:2000 for WB and 1:500–1:5000 for ELISA is commonly used for polyclonal antibodies.
