Biological Background: LNPEP Function and Localization
- LNPEP encodes leucyl-cystinyl aminopeptidase, also known as insulin-regulated membrane aminopeptidase (IRAP), oxytocinase (OTase), and placental leucine aminopeptidase (P-LAP).
- Function: Releases N-terminal amino acids (preferentially cysteine and leucine) from peptide hormones, thereby regulating their activity.
- Degrades oxytocin, vasopressin, and angiotensin III, playing a key role in maintaining homeostasis during pregnancy.
- May inactivate neuronal peptides in the brain; cleaves Met-enkephalin and dynorphin, and binds angiotensin IV, potentially acting as the angiotensin IV receptor in the brain.
- Subcellular localization: Cell membrane (signal-anchor) and secreted forms exist; contains a transmembrane helix.
- Tissue specificity: Highly expressed in placenta, heart, kidney, and small intestine; lower levels in brain neurons, skeletal muscle, spleen, liver, testes, and colon.
- Keywords: Metalloprotease, zinc-binding, glycoprotein, phosphoprotein, acetylated; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-110) of human LNPEP; this region may be conserved across species, consistent with observed cross-reactivity with mouse and rat.
- For WB, the full-length protein has a predicted molecular weight of ~117 kDa; ensure adequate transfer conditions and use appropriate molecular weight markers.
- For IHC-P, antigen retrieval conditions may require optimization; validation in relevant tissue samples (e.g., placenta, kidney) is recommended.
- In ELISA, the antibody may be suitable for capture or detection; perform titration experiments to determine optimal working concentration.
