Biological Background: Leiomodin-1 Function and Localization
- Leiomodin-1 (LMOD1), also known as 64 kDa autoantigen 1D or smooth muscle leiomodin, is an actin-binding protein encoded by the LMOD1 gene and belongs to the tropomodulin family.
- It mediates nucleation of actin filaments and is required for proper contractility of visceral smooth muscle cells. Related references: PMID:28292896
- LMOD1 localizes to the cytoplasm, specifically associated with myofibrils, sarcomeres, and the cytoskeleton in muscle cells.
- Tissue specificity studies show detection in lung vascular smooth muscle, thyroid, extraocular smooth muscle, heart, aorta, skeletal muscle, colon, urinary bladder, uterus, stomach, and small intestine. Related references: PMID:27144530, PMID:2026759, PMID:11318603
- The protein is associated with alternative splicing and phosphorylation, and recognized as a thyroid-associated ophthalmopathy autoantigen.
- Mutations in LMOD1 have been linked to disease variants such as megacystis-microcolon-intestinal hypoperistalsis syndrome (MMIHS), highlighting its role in smooth muscle function.
Experimental Guidance and Technical Tips
- For WB, the predicted molecular weight is 67 kDa; however, alternative splicing may produce additional bands. Validate with appropriate positive controls (e.g., smooth muscle lysates).
- IHC-P detection may require antigen retrieval optimization and careful selection of tissue sections rich in smooth muscle (e.g., colon, bladder).
- For ELISA, consider using the immunogen sequence (aa 29-187) as coating antigen to assess antibody specificity.
