Biological Background: LMBRD1 Function and Localization
- Lysosomal cobalamin transport escort protein LMBD1, also known as NESI, LMBR1 domain-containing protein 1, and nuclear export signal-interacting protein; encoded by the LMBRD1 gene (C6orf209).
- Lysosomal membrane chaperone required for the export of cobalamin (vitamin B12) from the lysosome to the cytosol, enabling its conversion to cofactors (PubMed:19136951). It targets the ABCD4 transporter from the endoplasmic reticulum to the lysosome (PubMed:27456980) and forms a complex with ABCD4 and cytoplasmic MMACHC to transport cobalamin across the lysosomal membrane (PubMed:25535791). - Acts as an adapter protein playing an important role in mediating and regulating the internalization of the insulin receptor (INSR) via clathrin-mediated endocytosis through its interaction with adapter protein complex 2 (By similarity).
- Essential for the initiation of gastrulation and early formation of mesoderm structures during embryogenesis (By similarity).
- May play a role in the assembly of hepatitis delta virus (HDV) (Microbial infection).
- Subcellular localization includes endoplasmic reticulum membrane, lysosome membrane, cell membrane, and clathrin-coated vesicles.
- Isoform 3 is expressed in liver.
- Additional features: alternative splicing, glycoprotein, phosphoprotein, transmembrane protein with nine predicted transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 220-300, a region within the protein; consider this when designing detection or competition experiments.
- For Western blotting, a band near 61 kDa (predicted) is expected; validate with appropriate positive control lysates from human or mouse cells.
- In immunofluorescence, co-staining with lysosomal markers can help confirm proper localization.
- As a polyclonal antibody, lot-to-lot variability should be monitored; consider pre-adsorption or affinity purification if high specificity is required.
- The antibody is listed for ELISA; optimize coating and detection conditions for your specific assay format.
