Biological Background: LIS1 Function and Localization

  • Platelet-activating factor acetylhydrolase IB subunit beta (LIS1, also known as PAFAH1B1, LIS-1, PAF-AH 45 kDa subunit) is the regulatory beta subunit of cytosolic type I PAF acetylhydrolase (PAF-AH (I)), an enzyme that inactivates platelet-activating factor (PAF) by hydrolyzing its acetyl group.
  • LIS1 positively regulates the minus-end directed microtubule motor protein dynein, enhancing dynein-mediated microtubule sliding and targeting dynein to microtubule plus ends. It is required for dynein recruitment to microtubule plus ends and BICD2-bound cargos. Related references: PMID:22956769
  • Essential for neuronal migration during brain development: LIS1 regulates nucleokinesis, the process whereby the nucleus translocates into the anterior process of migrating neurons, and is required for proper activation of Rho GTPases and actin polymerization at the leading edge of neurons.
  • Subcellular localization includes cytoplasm, cytoskeleton, centrosome, spindle, and nuclear membrane, reflecting its roles in microtubule-dependent processes such as maintenance of Golgi integrity and coupling of nucleus and centrosome.
  • Fairly ubiquitously expressed in both frontal and occipital areas of the brain, consistent with its central role in neurodevelopment.
  • Post-translational modifications including acetylation and phosphorylation may regulate LIS1 function, and mutations in the gene cause lissencephaly, a severe brain malformation.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 1-100 of human LIS1 (P43034). For Western blot, a band at approximately 47 kDa is expected. Validate band specificity using appropriate controls (e.g., knockdown or knockout cell lysates).
  • This antibody has been tested in WB, IF/ICC, IP, and ELISA. For immunoprecipitation, optimize antibody concentration and incubation conditions. For immunofluorescence, permeabilization and fixation methods should be evaluated for your specific sample.
  • Cross-reactivity with mouse and rat is reported, making it suitable for studies in these model organisms. However, sequence alignment of the immunogen region with mouse and rat LIS1 should be considered when interpreting results.
