Biological Background: Lipocalin-2/NGAL Function and Localization
- Lipocalin-2/NGAL (LCN2) is a 25 kDa secreted glycoprotein also known as siderocalin, p25, and oncogene 24p3.
- Functions as an iron-trafficking protein: binds iron via siderophores such as 2,3-DHBA and mediates iron delivery (holo-form) or removal (apo-form), influencing intracellular iron concentration. Related references: PMID:12453413, PMID:20581821, PMID:27780864
- Iron-free apo-LCN2 induces apoptosis by decreasing intracellular iron and upregulating proapoptotic BCL2L11/BIM.
- Key innate immunity component: sequesters bacterial siderophores (e.g., enterobactin) to limit iron availability for pathogens; also binds Mycobacterium tuberculosis siderophores. Related references: PMID:27780864, PMID:15642259, PMID:21978368
- Subcellular localization includes secreted, cytoplasmic granule lumen, and cytoplasmic vesicle lumen.
- Strongly expressed in neutrophils, bone marrow, and epithelial tissues including uterus, prostate, salivary gland, stomach, appendix, colon, trachea, lung, and kidney medullary tubules. Related references: PMID:7683678, PMID:8298140, PMID:9339356, PMID:30418175
- Post-translational modifications include glycosylation, disulfide bonds, and pyrrolidone carboxylic acid formation.
Experimental Guidance and Technical Tips
- The antibody is raised against recombinant human/mouse Lipocalin-2/NGAL, making it suitable for detecting both endogenous and overexpressed forms.
- For western blot (WB), a band at approximately 23 kDa is expected; consider using reducing conditions.
- For immunohistochemistry (IHC-P), antigen retrieval may be required; validate in your tissue system.
- For immunofluorescence (IF-P), optimize permeabilization and fixation protocols due to secreted and vesicular localization.
- As a polymorphic monoclonal antibody (PolymAb®), it offers consistency; still, validate in your specific assay conditions.
