Biological Background: Lipocalin-2/NGAL Function and Localization
- Lipocalin-2 (LCN2), also known as NGAL, p25, or 24p3, is a secreted glycoprotein of the lipocalin family that plays critical roles in iron trafficking, innate immunity, and apoptosis (UniProt P80188).
- Iron-trafficking protein involved in apoptosis, innate immunity, and renal development: iron-bound form (holo-24p3) delivers iron, while iron-free form (apo-24p3) chelates intracellular iron, leading to apoptosis via BCL2L11/BIM. Related references: PMID:12453413, PMID:20581821, PMID:27780864
- In innate immunity, limits bacterial proliferation by sequestering iron bound to microbial siderophores such as enterobactin, and can bind siderophores from M. tuberculosis. Related references: PMID:27780864, PMID:15642259, PMID:21978368
- Subcellular localization includes secretion, and presence in the lumen of cytoplasmic granules and cytoplasmic vesicles (UniProt). Post-translational modifications include glycosylation, disulfide bond formation, and a pyrrolidone carboxylic acid modification.
- Detected in neutrophils and expressed in tissues prone to microbial exposure, including bone marrow, uterus, prostate, salivary gland, stomach, appendix, colon, trachea, lung, and kidney medullary tubules; absent from small intestine and peripheral blood leukocytes. - The immunogen sequence (amino acids 1–100 of human Lipocalin-2) is conserved in mouse and rat, consistent with reported cross-reactivity; however, validate expression in the relevant sample system.
- For Western blot, the theoretical molecular weight is 23 kDa; optimize antibody dilution and consider using a positive control such as neutrophil lysates.
- For ELISA applications, this polyclonal antibody can serve as a capture or detection reagent; pairing with a monoclonal antibody may improve specificity.
