Biological Background: Lipocalin-2/NGAL Function and Localization

  • Lipocalin-2 (LCN2), also known as NGAL, p25, or 24p3, is a secreted glycoprotein of the lipocalin family that plays critical roles in iron trafficking, innate immunity, and apoptosis (UniProt P80188).
  • Iron-trafficking protein involved in apoptosis, innate immunity, and renal development: iron-bound form (holo-24p3) delivers iron, while iron-free form (apo-24p3) chelates intracellular iron, leading to apoptosis via BCL2L11/BIM. Related references: PMID:12453413, PMID:20581821, PMID:27780864
  • In innate immunity, limits bacterial proliferation by sequestering iron bound to microbial siderophores such as enterobactin, and can bind siderophores from M. tuberculosis. Related references: PMID:27780864, PMID:15642259, PMID:21978368
