Biological Background: Hepatic Triacylglycerol Lipase (LIPC) Function and Localization
- LIPC, also known as hepatic triacylglycerol lipase, lipase member C, lysophospholipase, or phospholipase A1, is a key enzyme in lipid metabolism highly expressed in the liver.
- It catalyzes the hydrolysis of triglycerides and phospholipids present in circulating plasma lipoproteins, including chylomicrons, IDL, LDL, and HDL, releasing free fatty acids and smaller lipoprotein particles. Related references: PMID:12032167, PMID:26193433, PMID:7592706
- The enzyme possesses lysophospholipase activity and shows greater binding affinity for neutral lipid substrates compared to phospholipid substrates.
- In native LDL, LIPC preferentially hydrolyzes phosphatidylcholine species with polyunsaturated fatty acids at the sn-2 position. Related references: PMID:26193433
- LIPC is a secreted glycoprotein with heparin-binding properties, predominantly localized to the extracellular space and associated with lipoproteins in plasma.
- It undergoes co- and post-translational modifications including signal peptide cleavage and glycosylation, and has been identified by direct protein sequencing and proteomics.
- Genetic variants in LIPC are linked to quantitative HDL cholesterol traits (HDLCQ12) and lipid metabolism disorders, highlighting its clinical relevance.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 280–499 of human LIPC, spanning part of the catalytic domain; the antibody is expected to detect full-length endogenous LIPC.
- For WB, expect a band around 56 kDa; validate using positive control lysates (e.g., HepG2 cells) and account for potential glycosylation shifts.
- In IF/ICC, use hepatocyte-derived cell lines and optimize permeabilization to detect secreted proteins; include negative controls.
- ELISA applications should be tested with recombinant protein standards and appropriate sample matrices; check for matrix effects.
- Confirm antibody specificity via knockdown or knockout controls if available, especially for quantitative assays.
