Biological Background: LINGO1 Function and Localization
- LINGO1 functions as a component of the Nogo receptor (NgR) signaling complex, mediating myelin-associated inhibition of axonal regeneration through RhoA activation. Related references: PMID:14966521, PMID:15694321
- LINGO1 is an important negative regulator of oligodendrocyte differentiation and axonal myelination. Related references: PMID:15895088
- In the developing cortex, LINGO1 acts with RTN4 and RTN4R to regulate neuronal precursor cell motility.
- LINGO1 is a single-pass transmembrane protein located at the cell membrane.
- Expression is restricted to the central nervous system, with highest levels in amygdala, hippocampus, thalamus, and cerebral cortex; it is upregulated in substantia nigra neurons from Parkinson disease patients.
- The protein contains leucine-rich repeats and an immunoglobulin-like domain, and undergoes alternative splicing, glycosylation, and phosphorylation.
- Disease-associated variants of LINGO1 are linked to intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 42-240 of human LINGO1, covering the extracellular domain; this may contribute to recognition of multiple isoforms and explain the observed cross-reactivity with mouse.
- For Western blot, the predicted molecular weight is approximately 70 kDa; consider validating detection in relevant CNS-derived cell lines or tissue lysates (e.g., human or mouse brain).
- This antibody can be used in ELISA applications; direct ELISA with recombinant LINGO1 protein as the coating antigen may be suitable for titer determination.
- Given its cross-reactivity with mouse, the antibody can be considered for studies using mouse models of CNS injury or demyelinating diseases.
