Biological Background: LIMS1 Function and Localization
- LIMS1, also known as PINCH-1 or renal carcinoma antigen NY-REN-48, is a LIM domain-containing protein that acts as a key adaptor within the ILK-PINCH-PARVIN (IPP) complex.
- It directly binds to F-actin and promotes F-actin bundling, which generates the mechanical force required for actin cytoskeleton reorganization and dynamic cell adhesion events such as cell spreading and migration.
- The protein is an essential component of focal adhesions and localizes to cell junctions and the cell membrane, where it mediates cell–matrix interactions.
- LIMS1 is expressed in most human tissues but is notably absent or expressed at very low levels in the brain.
- The protein contains multiple LIM zinc-binding domains that facilitate protein–protein interactions, and it undergoes post-translational modifications including acetylation.
- Alternative splicing of the LIMS1 gene produces multiple isoforms, which may contribute to functional diversity in different cellular contexts.
Experimental Guidance and Technical Tips
- The immunogen used to generate this rabbit polyclonal antibody covers amino acids 1–325 of human LIMS1; therefore, it is expected to recognize multiple isoforms and may be suitable for detecting endogenous LIMS1 in human, mouse, and rat cell lysates.
- For Western blot (WB), a band at approximately 37 kDa is predicted. Optimization of sample preparation and antibody dilution is recommended to minimize non-specific binding.
