Biological Background: LIMA1 Function and Localization
- Encoded by the LIMA1 gene (also known as EPLIN), alternative splicing produces isoforms alpha and beta; isoform alpha is predominantly expressed in epithelial cells.
- Actin-binding protein that regulates cytoskeleton dynamics by increasing stress fiber number and size, inhibiting membrane ruffling, preventing actin filament depolymerization, and bundling filaments while delaying nucleation and reducing branched filaments. - Acts as a negative regulator of primary cilium formation. Related references: PMID:32496561
- Plays a role in cholesterol homeostasis by influencing plasma cholesterol levels through regulation of intestinal cholesterol absorption; may function as a scaffold protein that recruits MYO5B to NPC1L1 at the plasma membrane.
- Localizes to multiple subcellular structures including the cytoplasm, focal adhesions, stress fibers, cell membrane, ruffles, and lamellipodia.
- Highly expressed in placenta, kidney, pancreas, prostate, ovary, spleen, and heart; also detected in lung, liver, brain, skeletal muscle, thymus, testis, and intestine but absent in leukocytes. Isoform alpha is abundant in epithelial cells from mammary gland and prostate.
- Post-translational modifications include acetylation and phosphorylation; contains a LIM domain and is classified as a metal-binding zinc protein.

- The immunogen corresponds to amino acids 560–759 of human LIMA1, a region that may yield antibodies recognizing both isoforms; consider sequence alignment with the mouse ortholog to assess cross-reactivity.
- For Western blot, the expected molecular weight is approximately 85 kDa; validate band specificity using appropriate controls and lysates from human or mouse cells.
- In immunohistochemistry (paraffin), test on human epithelial tissues (e.g., breast, prostate) where LIMA1 alpha is highly expressed; antigen retrieval conditions may require optimization.
- The polyclonal nature provides detection of multiple epitopes, making it suitable for sensitive ELISA applications; pairing with a monoclonal capture antibody is recommended for sandwich ELISA.
