Biological Background: LIM Kinase 1 (LIMK1) Function and Localization
- LIM kinase 1 (LIMK1) is a serine/threonine-protein kinase that governs actin filament dynamics by phosphorylating and inactivating cofilin (CFL1), cofilin-2 (CFL2), and destrin (DSTN), thereby stabilizing F-actin. Related references: PMID:11832213, PMID:15660133, PMID:16230460
- It functions downstream of Rho family GTPase pathways and is activated through phosphorylation by upstream kinases such as ROCK1, PAK1, and PAK4. Related references: PMID:10436159
- Beyond actin regulation, LIMK1 phosphorylates TPPP to promote microtubule disassembly and has been implicated in axonal outgrowth and brain development. Related references: PMID:18028908
- Subcellularly, LIMK1 localizes to the cytoplasm, nucleus, cytoskeleton, and lamellipodia, consistent with its role in cell motility and morphology.
- The protein is highly expressed in the nervous system, especially the cerebral cortex, and to a lesser extent in heart and skeletal muscle.
- Multiple protein bands (33, 68, 70, 72 kDa) detected by this antibody may represent different isoforms or post-translational modifications of LIMK1.
- Keywords associated with LIMK1 include kinase, LIM domain, zinc-binding, phosphoprotein, and its involvement in Williams-Beuren syndrome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (amino acids 630–647) of human LIMK1, which is conserved in mouse and rat, likely explaining the observed cross-reactivity.
- For Western Blot analysis, multiple bands may appear due to isoforms or cleavage products; the predicted UniProt molecular weight is approximately 72.6 kDa.
- In IHC-P experiments, consider optimizing antigen retrieval and validate staining in target tissues such as brain or heart where LIMK1 is enriched.
- For ELISA applications, suitable coating conditions and detection reagents should be determined empirically.
