Biological Background: LILRB4 Function and Localization

  • LILRB4 (Leukocyte immunoglobulin-like receptor subfamily B member 4) is a member of the leukocyte immunoglobulin-like receptor family, also known as ILT3, CD85K, or LIR-5, and functions as an inhibitory receptor involved in immune regulation.
  • It down-regulates immune responses and contributes to the development of immune tolerance. Related references: PMID:11875462
  • LILRB4 serves as a receptor for multiple ligands, including fibronectin (FN1), apolipoprotein E (APOE), and ALCAM/CD166. Related references: PMID:34089617, PMID:30333625, PMID:29263213
  • It inhibits FCGR1A/CD64-mediated monocyte activation by inducing phosphatase-dependent down-regulation of LCK, SYK, LAT, and ERK phosphorylation, leading to reduced TNF production, in part via FN1 engagement. Related references: PMID:19833736, PMID:34089617
  • LILRB4 inhibits T cell proliferation, induces anergy, suppresses differentiation of IFNG-producing CD8+ cytotoxic T cells, and enhances generation of CD8+ T suppressor cells. Related references: PMID:16493035, PMID:19833736, PMID:29263213
  • It up-regulates CD86 on dendritic cells, potentially influencing T cell co-stimulation. Related references: PMID:19860908
  • The receptor interferes with TNFRSF5/CD40 signaling and NF-kappa-B activation. Related references: PMID:11875462
  • LILRB4 is a transmembrane protein localized to the cell membrane and is expressed in monocytes, macrophages, dendritic cells, NK cells, B cells, and lung.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 22–259), so the antibody is expected to recognize the N-terminal region; consider this for detection of both membrane-bound and potential soluble isoforms.
  • For Western blot, a band near 49 kDa is anticipated, but post-translational modifications such as glycosylation may result in apparent molecular weight shifts.
  • Cross-reactivity with mouse and rat suggests epitope conservation across species; validation in the relevant model system is recommended.
  • For immunohistochemistry and immunofluorescence, appropriate fixation/permeabilization protocols should be optimized to preserve epitope accessibility.

CamelBio: Your One-Stop Sourcing Bridge

CamelBio provides diagnostic manufacturers and research laboratories with reliable access to IVD raw materials, including validated antibody pairs, optimized polyclonal antibodies, and bulk ancillary reagents for immuno-oncology and immune tolerance studies. Targeting LILRB4, an inhibitory immune receptor, our portfolio supports the development of assays exploring immune checkpoint modulation and antigen-presenting cell functionality. Whether you need rare-target antibodies or customized sourcing solutions, CamelBio bridges the gap from concept to clinic.
