Biological Background: LILRB2/CD85d/ILT4 Function and Localization
- LILRB2, also known as CD85d, ILT-4, LIR-2, and MIR-10, is a member of the leukocyte immunoglobulin-like receptor (LILR) family.
- It functions as a receptor for classical and non-classical MHC class I antigens, including HLA-A, HLA-B, HLA-C, HLA-G, and HLA-F, and is implicated in immune tolerance. Related references: PMID:11169396, PMID:12853576, PMID:16455647
- Binding of HLA-G in complex with beta-2 microglobulin and nonamer self-peptide triggers differentiation of regulatory T cells and myeloid-derived suppressor cells, crucial for maternal-fetal tolerance. Related references: PMID:16455647, PMID:20448110, PMID:27859042
- Competes with CD8A for MHC class I binding and negatively regulates FCGR1A-mediated phosphorylation and intracellular calcium mobilization. Related references: PMID:11875462, PMID:12853576, PMID:9548455
- Predominantly expressed on monocytes, with lower levels on dendritic cells, B cells, and NK cells; enriched in tolerogenic IL-10-producing dendritic cells and pregnancy-associated myeloid-derived suppressor cells. Related references: PMID:20448110, PMID:27859042
- Localizes to the cell membrane with a single-pass transmembrane domain and contains immunoglobulin-like domains characteristic of inhibitory receptors.
- Undergoes alternative splicing, alternative promoter usage, and alternative initiation, producing isoforms with distinct functional properties.
- Post-translational modifications include phosphorylation and glycosylation, which may regulate its signaling and surface expression.
Experimental Guidance and Technical Tips
- For Western blot, use reducing conditions and validate with positive controls such as LILRB2-expressing monocytic cell lines (e.g., THP-1) and negative controls (e.g., LILRB2-knockdown lysates).
- This antibody targets an intracellular epitope (aa 479-598); it is not suitable for live-cell flow cytometry without permeabilization.
- For ELISA, consider pairing with a capture antibody if developing a sandwich immunoassay, and optimize antigen coating and detection conditions.
- Inclusion of protease and phosphatase inhibitors in lysis buffers is recommended to preserve protein integrity and phosphorylation status.
