Biological Background: LIFR Function and Localization

  • LIFR is a transmembrane receptor that serves as the signal-transducing subunit for leukemia inhibitory factor (LIF) and related cytokines. It forms heterodimeric complexes with IL6ST/gp130 or participates in the ciliary neurotrophic factor receptor (CNTFR) complex together with CNTFR and IL6ST/gp130. Related references: PMID:11285233, PMID:11294841, PMID:1542794
  • Direct binding of LIF, oncostatin M (OSM), or cardiotrophin-1 (CTF1) triggers signaling. Additional cytokines such as ciliary neurotrophic factor (CNTF) and cardiotrophin-like cytokine factor 1 (CLCF1) require prior binding to the non-signaling receptor CNTFR to initiate signaling through LIFR. Related references: PMID:11285233, PMID:11294841, PMID:1542794
  • Ligand-induced heterodimerization activates associated JAK kinases, which phosphorylate the intracellular domains of IL6ST/gp130. This creates docking sites for STAT transcription factors, leading to their phosphorylation and activation. LIFR engages site 3 of ligand cytokines. Related references: PMID:11285233, PMID:11294841, PMID:9030543, PMID:36930708
  • A soluble or alternative form of LIFR can inhibit the biological activity of LIF by blocking its binding to membrane-bound receptors on target cells.
  • LIFR is primarily localized at the cell membrane; a secreted form also exists due to alternative splicing.
  • The protein contains a signal peptide, extracellular repeats, a transmembrane helix, and intracellular tyrosine residues subject to phosphorylation. It undergoes glycosylation and disulfide bond formation.
  • Mutations in LIFR are associated with disease phenotypes, and the gene is involved in chromosomal rearrangements.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 500–700 of human LIFR, a region within the extracellular domain, potentially recognizing both membrane-bound and secreted receptor forms.
  • For Western blotting, consider starting with a dilution range of 1:500–1:2,000 and validate using cell or tissue lysates known to express LIFR (predicted molecular weight ~124 kDa). Include appropriate negative controls.
  • For ELISA applications, titrate the antibody to determine the optimal working concentration and pair with a compatible detection system.
  • This antibody cross-reacts with mouse LIFR; validation in mouse-derived samples is recommended before use.
