- The alpha subunit is synthesized as a precursor with a signal peptide, which is cleaved to generate the mature secreted protein; it contains several conserved disulfide bonds and N-glycosylation sites critical for proper folding and hormone activity.
- According to UniProt, CGA is a secreted glycoprotein with known 3D structures and has been characterized by direct protein sequencing, confirming its role in endocrine regulation.
Experimental Guidance and Technical Tips
- The immunogen was designed using the mature peptide regions (aa 25-116) of both CGA and LHB, which excludes the signal peptide; this region covers the functional domains of the subunits.
- For Western blotting under reducing conditions, expect bands at approximately 13 kDa (CGA) and 15 kDa (LHB); consider using positive controls such as pituitary extract or recombinant heterodimer preparations.
- For immunohistochemistry (IHC-P), antigen retrieval (e.g., heat-induced epitope retrieval at pH 6 or 9) may be required to unmask the epitope; validate in relevant tissue samples known to express LH, such as pituitary or placental tissues.
- The antibody is applicable to ELISA; when setting up sandwich ELISA, pair with another antibody recognizing a distinct epitope on the same subunit or the cognate beta subunit to avoid steric hindrance.
