- LGR5 (Leucine-rich repeat-containing G protein-coupled receptor 5) is also known as GPR49, GPR67, and HG38, and belongs to the G protein-coupled receptor family with leucine-rich repeats in its extracellular domain.
- It functions as a receptor for R-spondins (RSPO1-4), potentiating the canonical Wnt signaling pathway by associating with phosphorylated LRP6 and frizzled receptors.
- Unlike classical GPCRs, LGR5 does not activate heterotrimeric G proteins to transduce signals; instead, it acts as a stem cell marker in the intestinal epithelium and hair follicle.
- The protein is localized to the cell membrane, Golgi apparatus, and trans-Golgi network membrane.
- LGR5 undergoes alternative splicing, glycosylation, and contains disulfide bonds, as indicated by its UniProt keywords.
