Biological Background: LGI1 Function and Localization

  • LGI1, or epitempin-1, is a secreted glycoprotein belonging to the leucine-rich repeat (LRR) superfamily. It is encoded by the LGI1 gene in humans, with a calculated molecular mass of approximately 63.8 kDa (UniProt: O95970).
  • It functions as a regulator of voltage-gated potassium (Kv) channels, specifically slowing the inactivation of channels composed of KCNA1, KCNA4, and KCNAB1 subunits by preventing KCNAB1-mediated channel closure (By similarity).
  • LGI1 acts as a ligand for the transmembrane protein ADAM22, positively regulating synaptic transmission mediated by AMPA-type glutamate receptors (By similarity).
  • Through the phosphatidylinositol 3-kinase/ERK signaling pathway, LGI1 suppresses the production of matrix metalloproteinases MMP1 and MMP3, indicating a role in extracellular matrix remodeling (PubMed:15047712). Related references: PMID:15047712
  • Subcellularly, LGI1 is present in the cytoplasm, endoplasmic reticulum, Golgi apparatus, and is also secreted and localized to synapses, consistent with its roles in both intracellular trafficking and intercellular signaling.
  • Tissue expression is predominantly neural, with highest levels in the temporal neocortex, parietal and frontal cortices, and putamen; lower in occipital cortex and hippocampus; and absent in the cerebellum (at protein level) (PubMed:16787412). Related references: PMID:16787412
  • Post-translational modifications include N-linked glycosylation, and alternative splicing generates multiple transcript variants, though their functional distinctions are not well characterized.
  • Mutations in LGI1 are associated with autosomal dominant lateral temporal lobe epilepsy (ADLTE) and intellectual disability, highlighting its critical role in neuronal function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 301-557 of human LGI1 (C-terminal region). This antibody is expected to recognize the C-terminal portion of the protein, potentially detecting both full-length and some truncated isoforms containing this region.
  • For Western blot analysis, a band at approximately 64 kDa is anticipated. It is advisable to validate molecular weight with a positive control lysate from human, mouse, or rat brain tissue, as cross-reactivity is reported for all three species.
  • For ELISA, the antibody can be used for direct or indirect detection of recombinant or native LGI1. Optimal working concentrations should be determined empirically.
  • Upon receipt, store the antibody at -20°C or as indicated on the vial for long-term stability, avoiding repeated freeze-thaw cycles.
