Biological Background: Leukosialin Function and Localization
- Leukosialin (SPN/CD43) is a predominant cell surface sialoprotein expressed on leukocytes, including thymocytes, T-lymphocytes, neutrophils, plasma cells, and myelomas.
- Regulates multiple T-cell functions such as activation, proliferation, differentiation, trafficking, and migration; facilitates lymph node homing via association with ERM proteins (EZR, RDX, MSN).
- Negatively regulates Th2 cell differentiation, promoting Th1 lineage commitment, and induces IFN-gamma expression in T-cells during TCR activation. Related references: PMID:18036228
- Prepares T-cells for cytokine sensing by inducing expression of IFNGR and IL4R, enhancing their signaling, and clustering IFNGR with TCR. Related references: PMID:24328034
- Functions as a major E-selectin ligand mediating Th17 cell rolling on activated vasculature during inflammation; also acts as a T-cell counter-receptor for SIGLEC1.
- Provides anti-apoptotic signals that promote cell survival.
- Localizes to the cell membrane, microvilli, uropodium, and also to the nucleus and PML bodies.
- Extensively modified by glycosylation and phosphorylation; identified as a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (aa 276-400) and may recognize both glycosylated and unglycosylated forms; consider this when interpreting band shifts.
- For Western blotting, a band near 40 kDa is expected, though extensive glycosylation can cause an apparent higher molecular weight. Validate in the relevant sample system.
- Optimize antibody dilutions for IHC-P and IF/ICC applications; antigen retrieval conditions should be tested empirically.
- Confirm reactivity in mouse and rat samples due to reported cross-reactivity; validate in the target tissue/cell type.
