Biological Background: LETMD1 Function and Localization

LETMD1 (LETM1 domain-containing protein 1) is also known as cervical cancer proto-oncogene 1 protein p40 (HCCR-1), HCCR2, and SLC55A3. It is a multi-pass membrane protein belonging to the LETM1 family.

Plays an essential role in mitochondrial structure and function, and is required for thermogenesis of brown adipocytes. In brown adipose tissue, it localizes to the nucleus and interacts with chromatin remodeler SMARCA4 to regulate thermogenic genes such as UCP1 (By similarity).

For Western blot, consider using a starting dilution of 1:500–1:2000 and validate with appropriate positive and negative controls. Endogenous LETMD1 expression may vary across tissues; use a high-expression tissue (e.g., kidney or liver) as positive control.

For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimization of antigen retrieval, fixation, and permeabilization is recommended. Include isotype controls to assess specificity.
