- LEO1 (RNA polymerase-associated protein LEO1, also known as replicative senescence down-regulated leo1-like protein) is an essential component of the PAF1 complex (PAF1C), a multifunctional transcriptional regulatory complex.
- PAF1C associates with RNA polymerase II via its CTD and is involved in transcriptional elongation, acting independently and synergistically with elongation factors such as TCEA1, DSIF, and HTATSF1.
- LEO1 is required for the transcription of Hox and Wnt target genes, connecting PAF1C to the Wnt signaling pathway.
- PAF1C plays a role in hematopoiesis and leukemogenesis by stimulating KMT2A/MLL1 transcriptional activity, particularly in the context of KMT2A/MLL1-rearranged oncoproteins (e.g., MLL-AF9, MLL-ENL).
- PAF1C regulates histone modifications, including monoubiquitination of histone H2B at Lys-120 (H2BK120ub1) by recruiting the RNF20/40 ubiquitin ligase complex, and subsequent methylation of histone H3 at Lys-4 (H3K4me3).
