Biological Background: Legumain Function and Localization
- Legumain (LGMN), also known as asparaginyl endopeptidase (AEP), is a lysosomal cysteine protease that exhibits strict specificity for hydrolysis of asparaginyl bonds and can slowly cleave aspartyl bonds under acidic conditions. Related references: PMID:23776206
- It plays a critical role in the processing of proteins for MHC class II antigen presentation within the lysosomal/endosomal system. Related references: PMID:9872320
- Legumain is also involved in MHC class I antigen presentation in cross-presenting dendritic cells by mediating cleavage and maturation of Perforin-2 (MPEG1), promoting antigen translocation in the cytosol (By similarity).
- Required for normal lysosomal protein degradation in renal proximal tubules and for degradation of internalized EGFR, thereby regulating cell proliferation (By similarity).
- The protein is ubiquitous but particularly abundant in kidney, heart, and placenta.
- Subcellularly localized to the lysosome.
- Contains disulfide bonds, is glycosylated, and is synthesized as a zymogen that requires proteolytic activation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 26–288 of human legumain, covering the mature protease domain; this design supports detection of both pro and active forms.
- For Western blotting, the predicted molecular weight is 49 kDa; consider that the proform may migrate at a higher molecular weight. Validate using positive control lysates from kidney or heart.
- In immunohistochemistry (IHC-P), antigen retrieval steps may be required. Tissues such as kidney or placenta, which express high levels of legumain, can serve as positive controls.
- For immunofluorescence (IF/ICC), expect a punctate, lysosomal staining pattern. Co-localization with lysosomal markers is recommended to confirm target specificity.
