Biological Background: LEF1 Function and Localization
- LEF1 is a nuclear transcription factor that binds DNA in a sequence-specific manner (PubMed:2010090). It plays a central role in the Wnt signaling pathway. Related references: PMID:2010090
- Upon association with CTNNB1 and EP300, LEF1 activates target gene transcription, while TLE family members (TLE1–4) act as repressors (PubMed:11266540). Related references: PMID:11266540
- LEF1 regulates T-cell receptor alpha enhancer function and is required for IL17A-expressing gamma-delta T-cell maturation (PubMed:19653274). Related references: PMID:19653274
- In pancreatic cancer, LEF1 activates MYC and CCND1, promoting proliferation; an isoform lacking the CTNNB1 binding domain antagonizes Wnt signaling.
- CTNNB1-independent functions include activation of fibronectin and repression of E-cadherin promoters, enhancing tumor cell migration and reducing aggregation.
- Localized predominantly to the nucleus, LEF1 is expressed in thymus and overexpressed in colon and pancreatic tumor biopsies.
- Post-translational modifications include phosphorylation and ubiquitination; it is a DNA-binding phosphoprotein implicated in ectodermal dysplasia.
- LEF1 also positively regulates odontoblast differentiation and may contribute to hair cell differentiation and follicle morphogenesis.
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted protein weight is ~44 kDa; validate band specificity using lysates from human and mouse cells. Consider using nuclear extracts to enrich LEF1 signal.
- In immunoprecipitation (IP), the recombinant immunogen (aa 1-300) targets the N-terminal region, so it may detect multiple LEF1 isoforms. Confirm capture efficiency with known interacting partners.
- For ELISA, optimize coating conditions and use recombinant protein as a standard to generate quantitative calibration curves.
- Species cross-reactivity has been demonstrated for human and mouse; for other species, sequence homology should be verified before use.
- Given the role in Wnt signaling, this antibody can be employed to probe pathway activation in cancer models; however, empirical validation in each experimental context is advised.
