Biological Background: LEF1 Function and Localization
- LEF1, also known as LEF-1, TCF1-alpha, TCF10, TCF7L3, and TCF1ALPHA, is a member of the TCF/LEF family of transcription factors. Isoforms lacking the CTNNB1 interaction domain may act as antagonists of Wnt signaling.
- LEF1 binds DNA in a sequence-specific manner and participates in the Wnt signaling pathway, activating transcription of target genes in the presence of CTNNB1 and EP300. Related references: PMID:2010090
- TLE1, TLE2, TLE3, and TLE4 repress LEF1-mediated transactivation. Related references: PMID:11266540
- LEF1 regulates T-cell receptor alpha enhancer function. Related references: PMID:19653274
- LEF1 localizes to the nucleus and is detected in thymus. It is highly expressed in colon cancer biopsies and cell lines, and expressed in several pancreatic tumors, with weak expression in normal pancreatic tissue.
- LEF1 is required for IL17A-expressing gamma-delta T-cell maturation, acts as a positive regulator in odontoblast differentiation and hair cell differentiation, and in pancreatic cancer it activates MYC and CCND1 expression to enhance proliferation, while also regulating fibronectin and E-cadherin to promote cell migration.
