Biological Background: LEF1 Function and Localization
- LEF1 (Lymphoid enhancer-binding factor 1) is a sequence-specific DNA-binding transcription factor that participates in the Wnt signaling pathway. Related references: PMID:2010090
- It activates target gene transcription in the presence of CTNNB1 and EP300; TLE family proteins (TLE1-4) repress LEF1/CTNNB1-mediated transactivation. Related references: PMID:11266540
- LEF1 regulates T-cell receptor alpha enhancer function and is required for IL17A-expressing γδ T-cell maturation via BLK modulation (by similarity). Related references: PMID:19653274
- An isoform lacking the CTNNB1 interaction domain may act as a Wnt signaling antagonist.
- In pancreatic cancer, LEF1 transcriptionally activates MYC and CCND1 to enhance proliferation, while also promoting migration by repressing E-cadherin and activating fibronectin in a CTNNB1-independent manner.
- Subcellular localization is predominantly nuclear.
- Tissue specificity: detected in thymus; highly expressed in colon and pancreatic cancer biopsies and cell lines; weakly expressed in normal pancreatic tissue. Isoforms 1 and 5 are detected in pancreatic cell lines.
- Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitin conjugation. Classified as a DNA-binding transcription factor with roles in Wnt signaling and ectodermal dysplasia.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 20–132 of human LEF1 (NP_057353.1), a region encompassing the N-terminal β-catenin binding domain; the polyclonal nature may allow recognition of full-length LEF1 and isoforms containing this region.
- For Western blot, a band near 44 kDa is expected; validate the band size in your sample system, as post-translational modifications may shift migration.
- The antibody cross-reacts with human, mouse, and rat; verify cross-reactivity in your specific matrix, as sequence homology in the immunogen region influences detection.
- Applications include WB, IP, IF-P (paraffin-embedded immunofluorescence), and ELISA; optimize dilutions and conditions empirically for each assay.
- Since LEF1 is a nuclear transcription factor, nuclear extraction or paraformaldehyde fixation may improve detection in IP and IF-P experiments.
