Biological Background: LCAT Function and Localization
- LCAT is a secreted enzyme that plays a central role in extracellular cholesterol metabolism by catalyzing the transfer of fatty acids from phosphatidylcholines (lecithins) to free cholesterol, generating cholesteryl esters and lysophosphatidylcholines on the surface of high-density lipoproteins (HDLs) and low-density lipoproteins (LDLs) (PubMed:10329423, PubMed:19065001, PubMed:26195816). Related references: PMID:10329423 PMID:19065001 PMID:26195816
- The enzyme exhibits substrate specificity for plasma 16:0-18:2 and 18:0-18:2 phosphatidylcholines (PubMed:8820107). Related references: PMID:8820107
- In the brain, LCAT is produced by astrocytes and esterifies cholesterol on nascent APOE-containing lipoproteins, influencing cerebrospinal fluid APOE and APOA1 levels and contributing to lipoprotein maturation (PubMed:10722751). Related references: PMID:10722751
- LCAT also functions as a platelet-activating factor (PAF) acetylhydrolase, catalyzing the hydrolysis of PAF to lyso-PAF and the transacetylation of PAF to 1-hexadecanoyl-sn-glycero-3-phosphocholine (PubMed:8016111). Related references: PMID:8016111
- Additionally, LCAT catalyzes the esterification of (24S)-hydroxycholesterol (cerebrosterol) to form 24(S)OH-cholesterol monoesters, potentially important in brain sterol metabolism (PubMed:24620755). Related references: PMID:24620755
- LCAT is a secreted glycoprotein with a signal peptide and contains disulfide bonds. It has a molecular weight of approximately 50 kDa (UniProt: 49578 Da; product specification: 50 kDa).
- Expression is highest in liver, brain, and testes, and the protein is detectable in blood plasma and cerebrospinal fluid.
- Mutations in LCAT are associated with familial LCAT deficiency and fish-eye disease, highlighting its critical role in lipoprotein metabolism.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to the N-terminal region (amino acids 1–100) of human LCAT. Verify that your target of interest contains this epitope.
- For western blot (WB) analysis, a band around 50 kDa is expected. Consider using a positive control such as human plasma or liver lysate, and optimize protein loading and antibody dilution for your system.
- The antibody is reported to cross-react with mouse and rat LCAT. If using the antibody for cross-species applications, validate specificity and sensitivity in the relevant sample system.
- For ELISA applications, this antibody may be used for quantitative detection of LCAT in biological samples. Include appropriate standards and optimize dilutions to ensure assay linearity and accuracy.
